[Pharmacological Actions]
Pharmacodynamics Florfenicol is a broad-spectrum amphenicol antibiotic that acts as a bacteriostatic agent, exerting its effect by binding to the 50S ribosomal subunit and inhibiting bacterial protein synthesis. It exhibits potent antibacterial activity against a wide range of Gram-positive and Gram-negative bacteria. Haemophilus parasuis, Pasteurella multocida, and Actinobacillus pleuropneumoniae are highly susceptible to florfenicol. In vitro, florfenicol displays antibacterial activity comparable to or even greater than that of thiamphenicol against many microorganisms; moreover, certain bacteria that have developed resistance to amphenicols through acetylation—such as Escherichia coli and Klebsiella pneumoniae—may still remain susceptible to florfenicol.
It is primarily indicated for bacterial diseases in pigs, chickens, and fish caused by susceptible pathogens, including respiratory diseases in cattle and pigs due to Haemophilus parasuis, Pasteurella multocida, and Actinobacillus pleuropneumoniae; typhoid and paratyphoid fever caused by Salmonella; fowl cholera, pullorum disease, and colibacillosis in chickens; as well as bacterial septicemia, enteritis, and red-skin disease in fish resulting from Pasteurella, Vibrio, Staphylococcus aureus, Aeromonas hydrophila, and Enterobacter species.
Pharmacokinetics Florfenicol is rapidly absorbed after oral administration, reaching therapeutic concentrations in the blood within approximately 1 hour and attaining peak plasma levels in 1 to 3 hours. Its bioavailability exceeds 80%. Florfenicol is widely distributed throughout the animal body and can cross the blood–brain barrier. It is primarily excreted unchanged in the urine, with a small fraction eliminated in the feces.
[Drug Interactions]
(1) Macrolides and lincosamides share the same target site as this product—binding to the 50S ribosomal subunit of bacteria—and may exhibit antagonistic effects when used in combination.
(2) It may antagonize the bactericidal activity of penicillins or aminoglycosides, but this has not yet been demonstrated in vivo in animals.
[Functions and Uses] An amide alcohol antibiotic. Used for infections caused by Pasteurella and Escherichia coli.
【Usage and Dosage】 Based on this product. For oral administration: for pigs and chickens, 0.1–0.15 g per kg body weight, twice daily for 3–5 consecutive days; for fish, 50–75 mg once daily for 3–5 consecutive days.
[Adverse Reactions] When used at doses higher than the recommended level, this product exhibits a certain immunosuppressive effect.
[Precautions]
(1) Chickens raised for egg production shall not be used during the laying period.
(2) Use with caution in breeding chickens. It exhibits embryotoxicity; use with caution in pregnant and lactating livestock.
(3) Do not use in animals during the vaccination period or with severe immune deficiency.
(4) In animals with renal insufficiency, the dosage should be appropriately reduced or the dosing interval extended.
