    [Pharmacological Actions]

    Pharmacodynamics Tiamulin is a pleuromutilin antibiotic that exhibits bactericidal activity against susceptible bacteria at high concentrations. It inhibits bacterial protein synthesis by binding to the 50S ribosomal subunit.

    Tiamulin exhibits potent antibacterial activity against Mycoplasma and Brachyspira hyodysenteriae, and also demonstrates good activity against most Gram-positive bacteria, including staphylococci and streptococci (except for Group D streptococci). It has some efficacy against Actinobacillus pleuropneumoniae, but its antibacterial activity against most Gram-negative bacteria is relatively weak.

    When combined with chlortetracycline at a ratio of 1:4, this product is effective in treating bacterial enteritis, bacterial pneumonia, and swine dysentery caused by Brachyspira in pigs. It also demonstrates significant therapeutic efficacy against pneumonia resulting from mixed infections involving Mycoplasma pneumoniae, Bordetella bronchiseptica, and Pasteurella multocida.

    Pharmacokinetics Oral administration in pigs results in good absorption. Following a single dose, the bioavailability is approximately 85%, with peak plasma concentrations reached within 2 to 4 hours. Once absorbed, the drug is widely distributed throughout the body, with the highest concentrations found in lung tissue. Tiamulin is extensively metabolized in vivo, yielding more than 20 metabolites, some of which exhibit antibacterial activity. Metabolites of tiamulin are primarily excreted in the feces via bile, with about 30% eliminated in the urine.

    [Drug Interactions]

    (1) Concurrent use with polyether antibiotics such as monensin, salinomycin, and methylsalinomycin may interfere with the metabolism of these polyether antibiotics, resulting in stunted growth, ataxia, paralysis, and even death.

    (2) When used in combination with other antibiotics that bind to the 50S subunit of bacterial ribosomes, such as macrolides and lincomycin, competitive binding at the same site may result in reduced therapeutic efficacy.

    [Functions and Uses] A truncated pleuromutilin antibiotic. It is primarily used for the prevention and treatment of chronic respiratory disease in chickens, mycoplasmal pneumonia and Actinobacillus pleuropneumonia in pigs, and also for swine dysentery and proliferative enteritis caused by spirochetes.

    【Usage and Dosage】 Based on this product. For mixed drinking: for pigs, 0.1–0.13 g per 1 L of water, administered continuously for 5 days; for chickens, 0.28–0.56 g per 1 L of water, administered continuously for 3 days.

    [Adverse Reactions] When used at the recommended dose, pigs may occasionally develop skin erythema. Overdose may cause transient salivation, vomiting, and central nervous system depression in pigs.

    (1) Concurrent use with polyether antibiotics such as monensin, salinomycin, and methylsalinomycin is prohibited.

    (2) Users should avoid contact between the medication and the eyes or skin.
